Buntanetap

Buntanetap (also known as Posiphen or ANVS-401; sometimes referred to informally as a “tab” or tablet/capsule) is an investigational oral small-molecule drug being developed by Annovis Bio for neurodegenerative diseases like Alzheimer’s disease (AD) and Parkinson’s disease (PD).10

What it is and how it works

  • It acts as a translational inhibitor of neurotoxic aggregating proteins (TINAPs). It reduces the production of multiple harmful proteins at once, including amyloid precursor protein (APP)/amyloid-beta, alpha-synuclein, tau, huntingtin, and TDP-43.17
  • This differs from most current therapies (e.g., antibodies targeting only one protein) by addressing several disease-related proteins via the iron response element (IRE) in their mRNA, inhibiting their translation without affecting normal proteins much.15
  • It’s taken orally (capsules/tablets), which is convenient compared to infusions.

Current development status (as of mid-2026)

  • Parkinson’s disease: Positive FDA notice to proceed with Phase 3; also has an open-label extension study for long-term safety/efficacy (up to 3 years, 30 mg daily).10
  • Alzheimer’s disease: In Phase 3 (pivotal trial ongoing); Phase 2/3 data showed safety and hints of cognitive/motor benefits.12
  • Well-tolerated in trials up to 80 mg daily, with good pharmacokinetics. Phase 2a showed improvements in motor function (PD) and cognition (AD).11

It is not yet approved by the FDA or other regulators — it’s still in clinical trials.

Potential benefits (from early data)

  • May slow disease progression by lowering toxic protein levels.
  • Shown promise in improving cognition and daily function in early studies.
  • Safer profile than some related compounds (e.g., lacks strong cholinesterase inhibition of its parent compound phenserine).
